Traditional Water

The single-cell suspension prepared using the Novel Bio Creation Automated Tissue Dissociator demonstrated comparable cell viability to the traditional water bath shaking method. However, the automated dissociator yielded significantly higher cell concentration and greater total cell numbers. These results validate the instrument's excellent performance in gentle yet efficient tissue digestion and highlight its advantage of high sample recovery rate.

Following dissociation, cells were captured using the NovelCyto platform, followed by library preparation and sequencing. Data analysis was performed with the CytoNavigator™ single-cell data analysis system, enabling detailed comparison between mouse bladder cancer subcutaneous tumor tissues processed by the Novel Bio Creation Automated Tissue Dissociator versus the traditional water bath shaking method.

Cell Quality Control:

• The Novel Bio Creation Automated Tissue Dissociator yielded 10,737 cells after sequencing, with a median genes per cell of 3,017.
• The traditional water bath shaking method yielded 10,729 cells, with a lower median genes per cell of 2,805.

This result demonstrates the advantage of the Novel Bio Creation Automated Tissue Dissociator in obtaining high-quality cells with superior gene expression data integrity.

Cell Type Quantification and Proportion Statistics:

Analysis of cell counts and proportions revealed significant advantages for parenchymal cell recovery:
The Novel Bio Creation Automated Tissue Dissociator yielded 142 Endothelial cells and 459 Fibroblasts.
The traditional water bath shaking method yielded only 64 Endothelial cells and 340 Fibroblasts.
This striking comparison clearly demonstrates the superior performance of the Novel Bio Creation Automated Tissue Dissociator in obtaining substantially higher numbers of parenchymal cells.
